What Is AOD-9604?

AOD-9604 is a synthetic peptide based on a small section of human growth hormone (HGH).

Researchers originally developed it in hopes of separating some of growth hormone’s effects on fat metabolism from its growth-promoting effects.

In other words, the idea was essentially: could researchers isolate the part associated with fat metabolism without producing all of the other effects of full growth hormone?

That made AOD-9604 particularly interesting as a potential obesity treatment in the early 2000s. 

Why Are Researchers Interested in It?

The biggest reason is fat metabolism and weight loss.

Early animal research suggested AOD-9604 could influence processes involved in breaking down stored fat, known as lipolysis, without producing the same IGF-1 effects expected from full growth hormone.

Those findings eventually led researchers to test AOD-9604 in people with obesity. 

AOD-9604 has also appeared in research involving cartilage and osteoarthritis, although this is a much less established area and should not be interpreted as evidence that it treats joint problems in humans. 

How Does It Work?

The proposed mechanism centers on fat metabolism.

Growth hormone affects many different systems throughout the body. AOD-9604 contains only a small fragment of the hormone and was designed to investigate whether some of its metabolic activity could be isolated.

Animal research suggested it could increase fat breakdown and fat oxidation without behaving like full growth hormone.

But importantly, how AOD-9604 behaves in animals doesn’t automatically tell us whether it produces meaningful fat loss in humans.

And that’s where the story gets interesting.

Unlike many research peptides, AOD-9604 actually progressed into human clinical studies.

Multiple randomized studies evaluated its safety, and researchers eventually conducted larger obesity trials. 

What Happened in the Weight-Loss Trials?

This is probably the most important part of the AOD-9604 story.

AOD-9604 was genuinely investigated as an obesity medication—it wasn’t simply a peptide that became popular online.

But the clinical development program ultimately didn’t demonstrate convincing weight-loss effectiveness.

FDA’s review of the available evidence notes that most identified studies failed to demonstrate a weight-reduction benefit compared with placebo. A large study involving 536 people with obesity did not find significant weight loss compared with placebo, and development of AOD-9604 for obesity was terminated. 

That’s a very important distinction:

AOD-9604 was studied for weight loss. That does not mean it was proven to work for weight loss.

What Do We Know?

There is legitimate science behind AOD-9604.

It is derived from human growth hormone, has demonstrated metabolic activity in preclinical research, and progressed far enough in development to be tested in people.

Human studies also provide some safety information. Published analysis of six randomized, double-blind, placebo-controlled trials reported that AOD-9604 was generally well tolerated within those studies and did not produce significant changes in IGF-1. 

However, tolerability and effectiveness are two different questions.

What Don’t We Know?

Current evidence does not establish that AOD-9604 reliably produces meaningful fat loss in humans.

Claims commonly seen online involving:

  • “Stubborn fat”
  • Belly-fat reduction
  • Rapid fat burning
  • Dramatically increased metabolism
  • Spot reduction
  • Muscle preservation during dieting

shouldn’t be treated as established human effects.

The larger obesity-development program simply didn’t produce the results necessary to establish AOD-9604 as an effective weight-loss drug. 

What About Joint & Cartilage Research?

AOD-9604 has also attracted interest outside weight management, particularly involving cartilage repair and osteoarthritis.

There is preclinical research in this area, but the evidence is substantially less developed than would be needed to establish a treatment.

FDA’s review noted that there was insufficient clinical information for it to evaluate proposed osteoporosis and osteoarthritis uses in compounded products. 

Research & Regulatory Status

AOD-9604 is not an FDA-approved medication.

In a 2024 review, FDA concluded that there was insufficient evidence supporting AOD-9604’s effectiveness for obesity and highlighted the failed larger obesity study. 

That makes its current popularity in the research-peptide and wellness space particularly interesting: the compound has a real pharmaceutical research history, but that history did not result in an approved weight-loss treatment.
